Orexin-B

Orexin-B (Hypocretin-2, OxB — Wakefulness Neuropeptide)
CognitiveFDA: Not approved — orexin agonists...

Overview

Discovered 1998. Orexin neuron loss is the established cause of narcolepsy type 1. Orexin receptor antagonists (suvorexant) are FDA-approved insomnia drugs, validating the pathway. Intranasal orexin-A restores wakefulness in narcoleptic primates.

Mechanism of Action

Endogenous neuropeptide binding OX1R/OX2R (10x selectivity for OX2R). Activates wake-promoting centers (locus coeruleus, TMN, raphe). Regulates wakefulness, arousal, appetite, reward. Orexin neuron loss causes narcolepsy type 1.
