P21

P21 (CNTF-Derived Neurogenic Peptide — Adamantylated Tetrapeptide)
CognitiveEmerging ResearchFDA: Not approved; preclinical

Overview

Preclinical: neurogenesis, cognitive enhancement, neuroprotection. Intranasal route exploits olfactory anatomy for rapid CNS delivery. No human clinical trials. CNTF literature provides mechanistic context.

Mechanism of Action

CNTF (ciliary neurotrophic factor) active domain derivative. Sequence: Ac-DGGLAG-NH2. N-terminal acetylation + C-terminal adamantylated glycine (AGly) enhances BBB penetration and metabolic stability. Same adamantane BBB strategy as Adamax. Promotes neurogenesis, hippocampal BDNF, synaptic plasticity. Intranasal: direct CNS via olfactory/trigeminal pathway — 15-min peak.
