FOXO4-DRI

FOXO4-DRI (Proxofim — Senolytic D-Retro-Inverso Peptide)
LongevityEmerging ResearchFDA: Not approved; preclinical research

Overview

2017 Cell paper (Baar MP et al.): FOXO4-DRI cleared senescent cells, restored fitness markers in aged mice. Major longevity interest. No human trials.

Mechanism of Action

Disrupts FOXO4–p53 protein interaction in senescent cells. p53 translocates to mitochondria → apoptosis in senescent cells. Spares healthy cells. First targeted senolytic peptide in mammals.
