NAD+

Nicotinamide Adenine Dinucleotide (NAD+)
LongevityExtensively StudiedFDA: Not approved as drug...

Overview

IV NAD+ widely used. SQ emerging. Oral bioavailability low. NMN/NR as oral precursors available. Clinical use for anti-aging, addiction, energy.

Mechanism of Action

Essential cellular coenzyme. Declines ~50% between ages 40–60. Cofactor for SIRT1–7 (sirtuins). Required for PARP-mediated DNA repair. Critical for mitochondrial ETC efficiency. Starting too high can cause insomnia/anxiety/fatigue.
