MOTS-c

MOTS-c (Mitochondrial Open Reading Frame of 12S rRNA-c)
LongevityExtensively StudiedFDA: Not approved; early research phase

Overview

Discovered 2015 (Lee et al., Cell Metabolism). Declining MOTS-c correlates with aging and metabolic disease. Exercise increases endogenous MOTS-c. Longevity extension in animal models.

Mechanism of Action

Encoded by mitochondrial genome (12S rRNA). Translocates to nucleus under metabolic stress. Activates AMPK. Improves insulin sensitivity. Reduces adipogenesis. Declines with age. ~12-fold increase in muscle with exercise.
